En tant qu’écophysiologiste, j’utilise pour mes recherches des approches intégratives allant de la molécule à l’animal entier afin de comprendre quels sont les mécanismes physiologiques sous-jacents aux trajectoires de vie des animaux. Plus précisément, je me concentre sur l’importance de la plasticité mitochondriale dans les réponses aux changements environnementaux. Je suis aussi intéressée de savoir si le métabolisme énergétique et le stress oxydant sont des causes proximales aux variations de trajectoires de vie et aux compromis physiologiques. La plupart de mes travaux utilisent les poissons comme modèle biologique.

oulos%22%7D%2C%7B%22creatorType%22%3A%22author%22%2C%22firstName%22%3A%22Neil%20B.%22%2C%22lastName%22%3A%22Metcalfe%22%7D%5D%2C%22abstractNote%22%3A%221.%20Many%20animals%20experience%20periods%20of%20food%20shortage%20in%20their%20natural%20environment.%20It%20has%20been%20hypothesised%20that%20the%20metabolic%20responses%20of%20animals%20to%20naturally-occurring%20periods%20of%20food%20deprivation%20may%20have%20long-term%20negative%20impacts%20on%20their%20subsequent%20life-history.%202.%20In%20particular%2C%20reductions%20in%20energy%20requirements%20in%20response%20to%20fasting%20may%20help%20preserve%20limited%20resources%20but%20potentially%20come%20at%20a%20cost%20of%20increased%20oxidative%20stress.%20However%2C%20little%20is%20known%20about%20this%20trade-off%20since%20studies%20of%20energy%20metabolism%20are%20generally%20conducted%20separately%20from%20those%20of%20oxidative%20stress.%203.%20Using%20a%20novel%20approach%20that%20combines%20measurements%20of%20mitochondrial%20function%20with%20in%20vivo%20levels%20of%20hydrogen%20peroxide%20%28H2O2%29%20in%20brown%20trout%20%28Salmo%20trutta%29%2C%20we%20show%20here%20that%20fasting%20induces%20energy%20savings%20in%20a%20highly%20metabolically%20active%20organ%20%28the%20liver%29%20but%20at%20the%20cost%20of%20a%20significant%20increase%20in%20H2O2%2C%20an%20important%20form%20of%20reactive%20oxygen%20species%20%28ROS%29.%204.%20After%20a%202-week%20period%20of%20fasting%2C%20brown%20trout%20reduced%20their%20whole-liver%20mitochondrial%20respiratory%20capacities%20%28state%203%2C%20state%204%20and%20cytochrome%20c%20oxidase%20activity%29%2C%20mainly%20due%20to%20reductions%20in%20liver%20size%20%28and%20hence%20the%20total%20mitochondrial%20content%29.%20This%20was%20compensated%20for%20at%20the%20level%20of%20the%20mitochondrion%2C%20with%20an%20increase%20in%20state%203%20respiration%20combined%20with%20a%20decrease%20in%20state%204%20respiration%2C%20suggesting%20a%20selective%20increase%20in%20the%20capacity%20to%20produce%20ATP%20without%20a%20concomitant%20increase%20in%20energy%20dissipated%20through%20proton%20leakage.%20However%2C%20the%20reduction%20in%20total%20hepatic%20metabolic%20capacity%20in%20fasted%20fish%20was%20associated%20with%20an%20almost%20two-fold%20increase%20in%20in%20vivo%20mitochondrial%20H2O2%20levels%20%28as%20measured%20by%20the%20MitoB%20probe%29.%205.%20The%20resulting%20increase%20in%20mitochondrial%20ROS%2C%20and%20hence%20potential%20risk%20of%20oxidative%20damage%2C%20provides%20mechanistic%20insight%20into%20the%20trade-off%20between%20the%20short-term%20energetic%20benefits%20of%20reducing%20metabolism%20in%20response%20to%20fasting%20and%20the%20potential%20long-term%20costs%20to%20subsequent%20life-history%20traits.%22%2C%22date%22%3A%22SEP%202018%22%2C%22language%22%3A%22English%22%2C%22DOI%22%3A%2210.1111%5C%2F1365-2435.13125%22%2C%22ISSN%22%3A%220269-8463%22%2C%22url%22%3A%22https%3A%5C%2F%5C%2Farchimer.ifremer.fr%5C%2Fdoc%5C%2F00449%5C%2F56022%5C%2F%22%2C%22collections%22%3A%5B%22348HK8H3%22%2C%22SC48RKDK%22%5D%2C%22dateModified%22%3A%222020-10-29T13%3A19%3A39Z%22%7D%7D%5D%7D
